面臨戰事即將爆發，將領們商議權宜之計時，Brabantio 對女兒 Desdemona 和 Othello 私婚之事感到憤怒，於是來到 Commander 面前請她主持公道。然而 Brabantio 對 Othello 的一句 “If thou hast eye to see; she hath deceived her father, and may thee” 揭開猜忌的序曲。
Othello 大敗敵軍後，在 Cyprus 街頭舉杯狂歡。在舞會時，Brabantio 先前的警告讓 Othello 對 Desdemona 和 Cassio 之間的互動心生猜疑。然而，黃湯下肚的 Cassio 因與 Montano 發生肢體衝突、遭到革職，進而轉向 Desdemona 求情復職。不料，Iago 和 Othello 目睹了一切的發生。
Iago 開始在 Othello 身旁縈繞不去，身處兩難的 Othello 卻無法自覺，其實 Iago 正是他內心黑暗面幻化出的心魔。
在地下室裡，Othello 內心猜忌的聲音所堆疊出來的幻想，使他對 Desdemona 和 Cassio 之間苟且的關係深信不疑，而 Desdemona 遺失的手帕，更是導致 Othello 邁向精神崩潰的關鍵。最後，結局會走向……
